1. Principle
The FSTest Ochratoxin Rapid Test Kit for Coffee is based onimmunochromato-graphic assay. The test cassette has a testing window for the observation of assay running and result reading. The testing window has an invisible T (test) line and a C (control) line before running the assay. When the treated sample was applied intothe sample hole on the device, the liquid will laterally flow throughthe surface of the test strip and react with the pre-coated recombinant antigens. If there are amount ochratoxin in the sample, The intensity of T line will be inhibited. Meanwhile, the Cline should always appear after a sample is applied, which indicates a valid result. By comparison the intensity between two lines, the device can indicate the presence of ochratoxin in the sample.

6. Sample preparation
6.1. Take an appropriate amount of coffee bean and grind it, (instant coffee is weighed directly), weigh 1 g of sample and add it into a 15 mL centrifuge tube.
6.2. Add 5 mL of extracting solution, re-cap the tube tightly, vortex for 3 mins to mix, then centrifuge at 4000 rpm for 3 mins.
6.3. Pipette 3 mL of supernatant into a 10 mL centrifuge tube, andevaporate the liquid in the sample with hot air at 60-65℃ by using nitrogen (air) dryer.
6.4. Add 500 μL of diluent solution, shake to mix.
7. Procedure & Steps
7.1. Before use, ensure the cassette is between 20 to 40℃.
7.2. Take the test cassette out of the bag and put it on a flat surface.
7.3 Pipette 100 μL of sample solution into a microwell, mix for 3-5 times with pipette, then stand for 3 mins.
7.4. Pipette 100 μL of sample solution into the sample hole of the test cassette.
7.5 Interpret the results after 8-10 mins. Results are invalid after 10 mins
