Description:
The FSTest Bacillus cereus count plate is a single use dehydrated culture medium which is ready-to-use. The plate contains nutrients, a cold-water-soluble gelling agent and a chromogenic indicator, allowing for the selective growth of Bacillus cereus from samples. The design facilitates colony growth and displays them in blue-green colony dots on the plate, providing shortened incubation time and enhanced visual interpretation.

Procedure & steps
1. Add 25 g or 25 mL sample to 225 mL sterilized phosphate buffer, or physiological saline diluent. Solid samples should be homogenized and liquid samples should be mixed to make a 1:10 dilution ratio sample solution. To make further serial dilutions, mix 1 mL diluted sample solution with 9 mL diluent.
2. Inoculation Open the package, place the FSTest Bacillus cereus count plates onto a sterile laboratory bench, clean bench, or biosafety cabinet, uncover the film on the top. Add 1 mL of sample solution onto the duplicated count plates, then recover the film gently and set for at least 10s to allow the solution to dry. Inoculate two count plates for each dilution ratio and one blank control needs to be included meanwhile. Generally, samples with 2 or 3 different dilution ratios need to be tested for each experiment.
Culture
Stack the count plates and put them back into the original plastic self-sealing bags (less than 12 pieces for each stack) and seal. Keep the plates in the incubator at 36±1°C for 15-24 hours with the film side upward.
Results Determination
There will be blue-green dots if the bacteria colonies grow on the aerobic count plate. Choose the count plates containing the 30-300 colonies and count these numbers. Multiply the numbers by corresponding dilution ratios to result in the total bacterial numbers in the original sample of each mL or g. It is recommended to double check positive results with other methods.
