Description:
FSTEST Shigella spp. Detecting Plate are sterile dehydrated culture medium prepared in a Petri dish, which is immediately ready to use. It provide the various nutrients and traceelements necessary for the growth of Shigella spp., along with inhibitors to suppress the growth of contaminants, lt also contains pH indicators and mixed dyes that react specifically with certain enzymes, which in turn hydrolyze substrates and release chromogenic groups.
Applications:
Intended to test Shigella spp. in food and food ingredients.
Pre-treatment steps
Prepare the 1:10 ratio solution by adding 25 g or 25 mL sample into the homogenization bag containing 225 mL sterile distilled water or saline and homogenize. According to the level of sample contamination and detection needs, a serial of dilutions can be prepared.
Procedure & Steps
1. Sample detection Uncover the upper film of the detection plate, pipette 1 ml of the sample solution to be tested and add it onto the detecting plate. Pick up the detecting plate and gently shake it until the sample is evenly spread across the surface of it and wait for 5 min for complete absorption. A serial of dilutions can be inoculated on duplicated plates. 2. Culture Stack together the detecting plates, incubate upside down at 36±1°C for 15-24 hrs.
Results Determination
Observe the colony characteristics on the detecting plate. Bacterial spots with white are considered positive for Shigella spp.. For samples with positive result, it is recommended to repeat the test with new detection plates or to perform necessary verificationusing other more reliable methods. If there is no bacterial growth in the detecting plate or colonies showing other colors, it can be determined as negative result.
