5. Test Procedure

STEP 1
Inoculate
Place plate on flat surface. Lift upper film and pipette 1.0 mL of liquid sample onto the center of the bottom film.
STEP 2
Incubate
Slowly roll down the top film to avoid bubbles. Stack plates (up to 20 high) film-side up and incubate at 36 ± 1 °C for 18-24 hours.
STEP 3
Interpret
Count red or reddish-brown colonies with a yellow acid halo using the built-in grid. Results can be counted manually or with an automated reader.

6. Performance & Validation

  • Testing Modes & Flexibility: Direct Method (for high bacterial load): Inoculate directly after a 1:10 dilution and serial dilutions.Enrichment Method (for extremely low bacterial load): Incubate statically in rapid enrichment broth at 36±1 ℃ for 7–8 hours prior to inoculation, effectively improving the detection rate.
