Step 1: Dilution- Homogenize 25 g/mL of sample with 225 mL diluent (1: 10), then perform serial dilutions (e.g., 1: 100) as needed.Purpose: Adjust sample concentration to ensure colony counts fall within the optimal range.
Step 2: Inoculation- Lift the top film, add 1 mL of sample to the center, and replace the film carefully to avoid air bubbles.Note: Air bubbles may interfere with colony growth and counting.
Step 3: Spread & Set- Use a spreader to distribute the sample evenly, then let the plate sit for 10 seconds to allow the gel to solidify.Function: Ensures uniform sample distribution and stable gel structure.
Step 4: Incubation-Stack plates horizontally (up to 20 high) and incubate at 36 ± 1°C for 18–24 hours (or 30 ± 1°C for dairy samples).Condition Adjustment: Temperature and time vary by sample type to optimize coliform growth.
Step 5: Counting- Count red colonies with gas bubbles (optimal range: 15–150 CFU), then multiply by the dilution factor to get the final concentration.Standard: 15–150 CFU ensures accurate and reliable counting results.
